Objectives

1. To study the protein translation machinery of Plasmodium
a. To investigate the dynamics of tRNA in Plasmodium
b. To characterize the tRNA recognition mechanisms in the parasite
c. To study gene translation in the apicoplast and cytosol of Plasmodium
2. To investigate the effects of known translation inhibitors in the malaria parasite
a. To characterize the activities of a large set of known and new inhibitors
b. To investigate their different effects upon the parasite by array techniques
c. To test their relative toxicity in a vertebrate model
3. To identify and characterize novel drug targets in Plasmodium falciparum
a. To obtain structural data for several Plasmodium aminoacyl-tRNA synthetases.
b. To obtain structural data for other components of the translation machinery.
c. To determine their evolutionary history and proximity to human orthologs
4. To generate new lead compounds and improve the methods to select them falciparum
a. To develope a new method of screening based on a positive selection scheme
b. To use our data to design novel potential inhibitory scaffolds
c. To synthesize several compound libraries and test their anti-malarial activity
